Would a light colour wash on a dark surface make a difference?

Sure - you'd create a reverse-contrast. What would you plan to do that to?

It would be the reverse of doing a dark wash on a light finish.
You could use it to simulate light colored dirt or sand or dust that 'ran' after a rainstorm.
I have also used some light color washes on dark green Soviet armor that I felt looked too 'black' and was happy with the results.
